โรงเรียนสาธิตมหาวิทยาลัยขอนแก่น เดิมชื่อ โรงเรียนสาธิตคณะศึกษาศาสตร์ มหาวิทยาลัยขอนแก่น สังกัดคณะศึกษาศาสตร์ มหาวิทยาลัยขอนแก่น ก่อตั้งขึ้นในปี พ.ศ. 2513 โดย ดร.สายสุรี จุติกุล คณบดีคนแรกของคณะศึกษาศาสตร์ เปิดสอนด้วยกัน 3 ระดับ คือ ระดับอนุบาล ระดับประถมศึกษา ระดับมัธยมศึกษา

โดยโรงเรียนสาธิตคณะศึกษาศาสตร์ มหาวิทยาลัยขอนแก่น เป็นแหล่งค้นคว้า วิจัย ทดลอง และสาธิตวิทยาการทางการศึกษาเพื่อให้เหมาะสมกับความแตกต่างกันระหว่างบุคคล มีคณาจารย์จากคณะศึกษาศาสตร์เป็นผู้ดำเนินการสอน โรงเรียนได้มีการรับนักเรียนโดยการทดสอบวัดความสามารถทางสติปัญญา ทั้งภายนอกและภายในมหาวิทยาลัยขอนแก่น ไม่มีนโยบายรับสวัสดิการ ตาบัญชา ชุมปิตาบัญชาชุปิ๊

ภายหลังมหาวิทยาลัยขอนแก่นได้ขยายหน่วยงาน ก่อตั้งคณะวิชาและสำนักงานต่าง ๆ เพิ่มมากขึ้น บุคลากรที่มาปฏิบัติหน้าที่ก็เพิ่มมากขึ้นตาม ทำให้บุคลากรของมหาวิทยาลัยมีปัญหาเรื่องที่เรียนของบุตร โรงเรียนสาธิตคณะศึกษาศาสตร์ มหาวิทยาลัยขอนแก่น ไม่สามารถสนองความต้องการนี้ได้ทั้งหมด เพราะต้องดำเนินตามวัตถุประสงค์และหลักการเดิมของการก่อตั้งโรงเรียนไว้ ในปี พ.ศ. 2524 รศ.นพ.กวี ทังสุบุตร อธิการบดีมหาวิทยาลัยขอนแก่นในขณะนั้น จึงมีดำริใหก่อตั้งโรงเรียนสวัสดิการขึ้น ใช้ชื่อว่า โรงเรียนสาธิตมอดินแดง มีสถานะเป็นโรงเรียนราษฎร์ สังกัดสำนักงานอธิการบดี เปิดสอน 3 ระดับเช่นเดียวกับโรงเรียนสาธิตคณะศึกษาศาสตร์ มหาวิทยาลัยขอนแก่น เมื่อดำเนินการได้ระยะหนึ่งโรงเรียนสาธิตมอดินแดงได้เกิดปัญหาขึ้นมากมาย โดยเฉพาะด้านการเงินและงบประมาณที่ไม่เพียงพอ บุคลากรขาดขวัญและกำลังใจ ลาออกเพื่อหางานที่มีความมั่นคงกว่า ทางผู้บริหาระดับคณะและมหาวิทยาลัยจึงได้ปรับเปลี่ยนวิธีการดำเนินการใหม่

พ.ศ. 2527 ได้มีการโอนโรงเรียนสาธิตมอดินแดงมาสังกัดคณะศึกษาศาสตร์ และได้รวมโรงเรียนสาธิตคณะศึกษาศาสตร์ มหาวิทยาลัยขอนแก่นและโรงเรียนสาธิตมอดินแดงเข้าด้วยกัน และยังคงให้ดำเนินการตามวัตถุประสงค์การจัดตั้งเดิม ภายใต้ชื่อ โรงเรียนสาธิตมหาวิทยาลัยขอนแก่น ประกอบด้วย โรงเรียนสาธิตมหาวิทยาลัยขอนแก่น (ศึกษาศาสตร์) เป็นโรงเรียนทดลองเพื่อศึกษาวิจัย และโรงเรียนสาธิตมหาวิทยาลัยขอนแก่น (มอดินแดง) เป็นโรงเรียนเพื่อสวัสดิการเพื่อบุคลากรของมหาวิทยาลัยขอนแก่น นอกจากนี้ยังให้ปรับเปลี่ยนเครื่องแต่งกายของโรงเรียนสาธิตมหาวิทยาลัยขอนแก่น (มอดินแดง) จากเดิมสีน้ำตาลแก่ มาเป็น สีเทาแก่ ให้เหมือนเครื่องแบบนักเรียนโรงเรียนสาธิตมหาวิทยาลัยขอนแก่น (ศึกษาศาสตร์)

พ.ศ. 2532 รศ.นพ.นพดล ทองโสภิต อธิการบดี และ รศ.ดร.แก้วตา คณะวรรณ คณบดีคณะศึกษาศาสตร์ ขณะนั้น มีนโยบายให้ย้ายนักเรียนโรงเรียนสาธิตฯ (มอดินแดง) มารวมกันที่โรงรียนสาธิตฯ (ศึกษาศาสตร์) ทั้งหมด เพื่อความเป็นเอกภาพ แต่ทำได้เพียงรับนักเรียนระดับอนุบาล 1 ของโรงเรียนสาธิตฯ (มอดินแดง) มาเรียนรวมกันที่โรงเรียนสาธิตฯ (ศึกษาศาสตร์) เป็นปีแรกเท่านั้น ทำให้โรงเรียนสาธิตมหาวิทยาลัยขอนแก่น (ศึกษาศาสตร์) ระดับอนุบาล เปลี่ยนมาเป็น โรงเรียนสาธิตมหาวิทยาลัยขอนแก่น ระดับอนุบาล

พ.ศ. 2542 สภามหาวิทยาลัย ได้มอบหมายให้คณบดีคณะศึกษาศาสตร์ ปรับแก้โครงสร้างเชิงบริหารของโรงเรียนสาธิตทั้ง 2 แห่ง ให้เป็นเอกภาพโดยให้มีผู้อำนวยการคนเดียวกัน รวมทั้งใช้นโยบายการจัดการเป็นอันเดียวกัน เพื่อลดช่องว่างระหว่างความรู้สึกเปรียบเทียบของผู้ปกครองที่ต้องการความทัดเทียม จึงการยกเลิกข้อบังคับมหาวิทยาลัยขอนแก่นว่าด้วยโรงเรียนสาธิตมหาวิทยาลัยขอนแก่น พ.ศ. 2534 โดยใช้ข้อบังคับฯ ว่าด้วยโรงเรียนสาธิตมหาวิทยาลัยขอนแก่น พ.ศ. 2544 และระเบียบว่าด้วยโรงเรียนสาธิตมหาวิทยาลัยขอนแก่น พ.ศ. 2546 แทน

พ.ศ. 2545 ได้มีการจัดตั้งศูนย์วิจัยและพัฒนาการจัดการศึกษาพิเศษแบบเรียมรวมสำหรับเด็กออทิสติก โรงเรียนสาธิตมหาวิทยาลัยขอนแก่น เพื่อพัฒนาหารูปแบบการพัฒนาเด็กออทิสติกให้เรียนรวมกับเด็กปกติได้

ปัจจุบันโรงเรียนสาธิตมหาวิทยาลัยขอนแก่น เป็นหน่วยงานเทียบเท่าภาควิชา สังกัดคณะศึกษาศาสตร์ มหาวิทยาลัยขอนแก่น ตั้งอยู่ 2 แห่งภายในมหาวิทยาลัยขอนแก่นได้แก่
