3.3. Comparison of PyOx and GOx in the bioactive layer
In order to investigate the advantageous of using PyOx in the
bioactive layer instead of GOx, another bi-enzymatic system was
prepared based on AG/GOx/CHIT in which GOx activity was equal to
PyOx which was 9.0 U and maltose concentration was estimated using
AG/GOx/CHIT biosensor at the same working conditions. Linear
range for maltose was found to be as 0.25 to 2.0 mM with y=
0.488x+0.044 (R2=0.993, x and y show concentration in mM and
current density as μA/cm2, respectively). While similar linear ranges
were obtained for both systems, approximately two times lower slope
value was observed by AG/GOx/CHIT in compared with AG/PyOx/CHI
in which this value was calculated as 0.844. This is an expected result
and due to from the lack of anomer specificity of PyOx so the both
β- and α-forms of glucose could be used as the substrate by this
enzyme unlike GOx.
